<p>超滤是一种膜分离技术， 它的特点是使用不对称多孔膜， 根据分子的大小来分离溶液中的大分子物质与小分子物质。 超滤尤其适用于大分子溶液的浓缩、 不同种类分子的纯化以及溶剂交换等。 超滤法是一种温和的、 非变性的物理方法， 比其它分离方法具有效率更高、 更灵活的优点。</p>
<h5><strong>超滤法的典型应用</strong></h5>
<ul>
<li>对蛋自质、 酶、 DNA、 单克隆抗体、 免疫球蛋白进行浓缩和脱盐</li>
<li>药物、 激素分析</li>
<li>从PCR扩增的DNA中去除引物</li>
<li>去除标记的氨基酸和核苷酸</li>
<li>HPLC样品制备</li>
<li>样品的除蛋白</li>
<li>从生物体液、 发酵肉汤培养基中纯化抗生素、 激素、 药物</li>
<li>从细胞悬液、 裂解液中回收生物分子对蛋白质、 酶、 细胞、 DNA、 生物分子、 抗体和免疫球蛋白进行常规意义的实脸室浓缩和脱盐</li>
<li>哺乳动物细胞的收集</li>
<li>清洗细胞、 纯化病毒、 去除细胞碎片、 除热原</li>
</ul>
<p>Sartorius提供了用于超滤的多种操作方式， 选择范围非常广泛。 选用何种操作方式主要根据样品处理量、 实验室现有没备、 滤过速度、 用户的实验要求等综合指标来决定。</p>
<h6><strong>离心</strong></h6>
<p>(处理最从100ul到l00ml)<br />
通过离心力， 使溶液中的小分子溶质和溶剂透过超滤膜， 被收集滤过液收集管中， 而大分子溶质则被超滤膜截留在样品浓缩管中。该方式的特点是操作简便， 滤过速度快。Sartonus提供了10种规格的浓缩离心管， 处理量从100ul到l00ml</p>
<h6><strong>加压离心</strong></h6>
<p>(处理量从5ml到50ml)<br />
加压离心是Sartorius公司特有的操作方式， 将浓缩离心管加压后再放入离心机里离心， 比单独离心快30-50%， 是最快的浓缩方式。Vivaspin 20和Vivacell 70也可以按照这种方式进行操作。</p>
<p><strong>加压</strong><br />
(处理量从5ml到250ml)<br />
用压缩空气或惰性气体作为超滤的动力， 超滤的同时通过振荡来避免浓差极化， 提高超滤速度。 Vivaspin 20、 Vivacell 70、 Vivacell 100和Vivacell 250适用于这种操作方式， 加压后静置即可完成超滤。 当需要快速超滤时， 可在加压后放在摇床上， 边振荡边超滤。</p>
<p><strong>切向流</strong><br />
(处理量从1OOml到5l以上)<br />
使用蠕动泵送液， 溶液沿与超滤膜平行的方向回旋流动， 可避免浓差极化， 提高超滤速度。Vivaflow 50和Vivaflow 200是专为这种操作方式设计的。</p>
<p><strong>溶剂吸附</strong><strong><br />
</strong>(处理量从1ml到20ml)<br />
该方式在超滤膜后加纤维素吸附垫， 以吸附力作为超滤的动力， 不需要任何附加设备。 溶剂和小分子溶质在吸附力的作用下透过超滤膜， 而大分子物质则被截留浓缩。 Vivapore是专用于这种操作方式的， 有5种规格可供选择， 样品处理量从Iml到20ml。</p>
